If you any of you guys are using Mac OS X might I suggest AudioFinder. I use it and it made culling through a library of over 1 million samples brain dead simple. It has saved me countless hours...

I don't have a huge library of samples and don't use a mac (yet, at least) - so I can't really add to this discussion, but I know a lot of people do have huge libraries out there...
